More than five million people in the US live with B cell-driven autoimmune disease, many diagnosed young and facing a lifetime of treatment. These diseases arise from complex, interconnected B cell biology. Single-target therapies leave many aspects of patients’ conditions unaddressed, and even emerging multi-target approaches often come with burdensome dosing regimens underscoring the need for treatments that combine broader target coverage with more convenient dosing.
Boulevard’s approach is grounded in a deep understanding of B cell disease biology, the identification of key mechanisms and targets that drive autoimmune disease pathology, and the ability to engineer multi-specific antibodies to address those drivers, with the potential to modify disease at its source.
IgA nephropathy: Silent, serious, underserved
IgA nephropathy is a chronic disease that begins in early adulthood and advances silently toward kidney failure. While emerging approaches address the underlying biology more precisely than in the past, they still require burdensome dosing regimens that weigh on patients across decades of treatment, impacting their quality of life.
BLVD101: Proven biology, designed for a lifetime
BLVD101 (formerly DFX-1092) is a dual BAFF/APRIL-targeting bispecific antibody designed to give patients more freedom and a better quality of life with dosing as infrequently as once every twelve weeks. Clinical data from dual BAFF/APRIL agents suggest that blocking both targets may be able to slow IgAN disease progression, drive reductions in proteinuria and hematuria, and reduce disease burden across renal and systemic autoimmunity. Beyond IgAN, dual BAFF/APRIL inhibition has the potential to serve as a backbone therapy across B cell-driven autoimmune diseases, including systemic lupus erythematosus (SLE) and Sjögren’s disease.
BLVD101 is being evaluated in an ongoing Phase 1 single-ascending dose study in healthy volunteers as a potential treatment for IgAN.
BLVD201: Compact design, deeper penetration
BLVD201 is a trispecific CD19/BCMA/CD3 T cell engager designed to reset the immune system at the root of disease, a potentially curative approach for severe B cell-driven autoimmune disease. Its compact design is built to penetrate deeply into tissue, enabling a robust response at a low dose. By engaging both CD19 and BCMA at once, it aims to clear antibody-producing cells and those that replenish them, potentially offering some patients a lifetime free from treatment.
BLVD201 is currently in IND-enabling studies.
BLVD301 is directed at a novel target combination in B cell-driven autoimmune disease and is supported by established genetic evidence.
BLVD301 was nominated as a development candidate in June 2026.
